Third of four videos on the discovery of DNA (third year high school program) Griffith's experiment and the subsequent one by Avery and collaborators had demonstrated with good certainty that it was DNA and not proteins that was the molecule responsible for genetic information. The resistance to this demonstration was difficult to dismantle, until the experiment by Hershey and Chase described in this video. The main objective of Hershey and Chase was therefore to determine which component of viruses, proteins or deoxyribonucleic acid (DNA), was responsible for transmitting genetic information to host cells. The two scientists managed to resolve the dilemma of whether the genetic material was made up of proteins or DNA. They used a very refined approach, using T2 Bacteriophages: viruses, which infect Escherichia coli bacteria and are made up of a protein capsid that wraps the genetic material (DNA). So they are only made up of DNA and proteins. To trace the DNA they used radioactive Phosphorus (³²P) and to trace the proteins they used radioactive Sulphur (³⁵S).
